Scarlett Johansson wouldbe a good choice for Samus. If I had the ability when I was old enough, I would hands down choose Dakota Fanning.
But Metroid is an E10+ - T rated franchise, so the possibilities of a Metroid movie being rated R are 0%, since Nintendo wants to attract just about everyone. Leave the grittiness to XBox. It's extremely likely a movie would be PG-13, especially since the best-selling films are usually PG-13. Titanic, Avatar, Transformers, LOTR, Skyfall, Dark Knight, Fast and the Furious... Metroid is in the hands of people like Shigeru Miyamoto, people who are not known for violence or sex. It's PG-13.

Scarlett Johansson would be a good choice to play Samus. the only 2 things she needs to fit the role would be dying her hair blonde and putting on some muscle, and try and get decent six-pack abs.
