Ralph is my favorite too. There was a hint mid way in the movie where the intent is to collect blood with that mutagen. If there is a sequel another Ralph would be made to stay true to the video game.

Not sure if heads could be regenerated anywhere in fiction. It seems like it breaks a rule.
Right now the only way possible is someone could regenerate another Ralph from the mutageon in his blood.

There are plenty of characters in fiction whom can regrow heads, quite a few (such as the Hulk) can come back from total obliteration to varying levels of speed. Still though, it may be kind of ridiculous given the film's semi-realistic tone.
